唐菖蒲的愈伤组织诱导和植株再生

摘 要:

以唐菖蒲品种 ‘Advanced red’ 的籽球、花柱、子房和花丝为外植体, 研究了不同种类和浓度生长调节物质以及籽球 的不同接种方式对愈伤组织诱导和植株再生的影响。结果表明: 唐菖蒲籽球的中部切片为最佳外植体, 倒接(形态学下端向 上)是最佳接种方式, 诱导非胚性(NEC)和胚性(EC)愈伤组织形成的最佳诱导培养基分别为MS+6 mg•L-1 NAA 和MS+6 mg•L-1 TDZ, 诱导率分别达 65.53% 和 71.88%。NEC 和 EC 均能分化不定芽, 分化率分别达 88.89% 和 61.11%。最佳生根培养基上 的生根率为87.50%。

Callus Induction and Plant Regeneration of Gladiolus hybridus Hort.

Abstract:

Effects of several plant growth regulating substances in different concentrations and different cormlet inoculation methods on callus induction and plant regeneration were investigated with cormlets, styles, ovary and stamens of Gladiolus hybridus cv. ‘Advanced red’ as explants. The results showed that the optimal explants were central sections of cromlets, the optimal inculation method was inverse inoculation. The optimal induction mediums for NEC and EC induction were MS+6 mg·L-1 NAA and MS+6 mg·L-1 TDZ with the induction rates of 65.53% and 71.88%, respectively. Calli from NEC and EC could differentiate adventitious shoots and the differentiation rates were 88.89% and 61.11%, respectively. Rooting rate on the optimal rooting medium was 87.50%.
